Ubikvitin-proteinska ligaza (EC 6.3.2.19, ubikvitin-aktivirajući enzim) je enzim sa sistematskim imenom ubikvitin:protein-lizin N-ligaza (formira AMP).[1][2][3][4] Ovaj enzim katalizuje sledeću hemijsku reakciju

ATP + ubikvitin + protein lizin {\displaystyle \rightleftharpoons } AMP + difosfat + protein N-ubikvitilizin

Ubikvitin je vezan za ovaj protein peptidnom vezom između C-terminalnog glicina ubikvitina i epsilon-amino grupa lizinskih ostataka proteina.
